Riding the Cable Car from Sewers to NEST: All previous areas (Raccoon City Police Department, Underground Facility, Sewers) become permanently inaccessible for the rest of that run.
Ada Wong's gameplay segment in Leon's campaign: Missable achievement "One Slick Super-spy" becomes impossible if failed or completed using weapons once control shifts back to Leon.
Sherry Birkin's gameplay segment in Claire's campaign: Missable achievement "Young Escapee" becomes impossible if the puzzle room is not escaped within 60 seconds once the cutscene triggers.
Boarding the escape train / elevator in NEST (Laboratory): Final point of no return for the entire facility; all uncollected items, files, and Mr. Raccoon figures in NEST become inaccessible.

Recommended order

Step 1Playthrough 1 - Leon Standard (Scenario A)

Focus on enjoying the story, learning map layouts, unlocking all safe and dial locks, destroying Mr. Raccoon figures, and gathering files. Complete Leon-specific achievements (such as unlocking Ada's hacking sequence without weapons).
Step 2Playthrough 2 - Claire Standard (Scenario B / 2nd Run)

Experience the alternate scenario, collect Claire-specific files and Mr. Raccoon figures, finish Sherry's Orphanage sequence within the time limit, and defeat the true final boss.
Step 3Playthrough 3 - Assisted Mode / Cleanup

Complete remaining miscellaneous achievements, such as "Frugalist" (no recovery items), "Minimalist" (never open the item box), and "A Small Carbon Footprint" (under 14,000 steps).
Don't miss in this step
- Frugalist: Complete an entire story campaign from start to finish without consuming any first aid sprays or herbs.
- A Small Carbon Footprint: Complete an entire story playthrough taking 14,000 steps or fewer, which can be tracked in the Records menu.
- Minimalist: Complete an entire story playthrough without interacting with or opening the item storage box even once.
Step 4Playthrough 4 - Leon Hardcore (Scenario A or B)

Complete the campaign on Hardcore difficulty to earn S ranks and unlock infinite weapons, while earning the Hardcore completion achievements.
Step 5Playthrough 5 - Claire Hardcore (Scenario A or B)

Complete Claire's campaign on Hardcore difficulty for her respective S rank and Hardcore completion achievements.
Step 6The 4th Survivor

Complete the unlocked bonus mode to earn the "Grim Reaper" achievement.
