Defeating Elder Jinchi in Chapter 1: Wandering Wight permanently despawns if not defeated first, locking out its spirit and achievement requirements for that playthrough.
Defeating Yellowbrow in Chapter 3: Locks the Pagoda Realm ward prisoner quest if all four Lantern Warden Captain spirits were not gathered, locking out the curio reward for that cycle.
Defeating Duskveil in Chapter 4 Secret Area: Permanently despawns Daoist Mi, locking his quest, transformation spell, and associated achievements for that playthrough.
Breaking the fourth Purple Altar and defeating Hundred-Eyed Daoist Master in Chapter 4: Locks the Supreme Inspector altar encounter questline if not finished prior to the chapter climax.
Defeating the final boss in Chapter 6 without visiting Mount Mei: Locks you out of the Secret True Ending and Erlang Shen's specific weapon/spirit unlocks for that playthrough.

Recommended order

Step 1

Start Playthrough 1 with Chapter 1 (Black Wind Mountain), ensuring you ring all three bells to defeat Elder Jinchi before defeating Black Bear Guai, and defeat the Wandering Wight before ringing the third bell.
Step 2

Progress through Chapter 2 (Yellow Wind Ridge), complete the Drunken Boar and Man-in-Stone questlines, defeat Tiger Vanguard, unlock the Secret: Kingdom of Sahali, and defeat Fuban before finishing Yellow Wind Sage.
Step 3

Explore Chapter 3 (The New West), ensuring you interact with the Fox spirit, complete the prisoner quest in Pagoda Realm by killing the four Captains before Yellowbrow, and finish Chen Loong's quest to unlock the Painted Realm hub.
Step 4

Navigate Chapter 4 (The Webbed Hollow), ensuring you defeat the Venom Daoist twice to unlock Secret: Purple Cloud Mountain, eliminate the Daoist Mi questline boss before Duskveil, peel all four Purple Altars before the final boss, and defeat Hundred-Eyed Daoist Master.
Step 5

Conquer Chapter 5 (Flaming Mountains), complete the Five Element Carts questline to enter Secret: Bishui Cave, acquire all Bull King iron materials, defeat Bishui Golden-Eyed Beast, and finish the chapter.
Step 6

Travel to Chapter 6 (Mount Huaguo), collect all Mythical Sun Wukong armor pieces, defeat the Giant Shigandang, unlock the secret Mount Mei area via the Great Pagoda to defeat Erlang Shen, and then defeat the final boss to achieve the true ending.
Step 7

Clean up remaining accessible spirits, gourds, soaks, skandhas, and meditation spots across all chapters before starting New Game+.
Step 8

Start New Game+ to acquire duplicate unique materials (such as Bull King's Iron Horns), craft all remaining weapons and armor sets, unlock remaining transformation spells, and collect the final achievements.
